What Is HPLC?

High-Performance Liquid Chromatography (HPLC) is an analytical technique used to separate, identify, and quantify components in a mixture. It is one of the most widely used methods in analytical chemistry and is a standard tool for assessing the purity of research peptides.

How Does HPLC Work?

In HPLC analysis, a sample is dissolved in a solvent and injected into a column packed with a stationary phase material. A mobile phase (liquid solvent) is pumped through the column under high pressure, carrying the sample components with it. Different components interact with the stationary phase to varying degrees, causing them to travel through the column at different rates and separate from one another.

As each component exits the column, it passes through a detector — commonly a UV detector — which generates a signal proportional to the amount of that component present. The result is a chromatogram showing peaks that correspond to individual components in the sample.

How Is HPLC Used for Peptide Purity Testing?

For research peptides, HPLC is used to determine purity by measuring the proportion of the target peptide peak relative to all detected peaks in the chromatogram. Purity is typically expressed as a percentage. A peptide with 98% HPLC purity, for example, means that 98% of the detected material corresponds to the target compound.

Why Is HPLC Important for Research Peptides?

HPLC purity data provides researchers with confidence that the compound they are working with is predominantly the intended peptide, with minimal impurities that could interfere with experimental results. It is a key component of a Certificate of Analysis and is considered a standard quality indicator in the research peptide industry.
