What Is LC-MS?

LC-MS stands for Liquid Chromatography–Mass Spectrometry. It is an analytical technique that combines the separation capabilities of liquid chromatography (LC) with the detection and identification capabilities of mass spectrometry (MS). LC-MS is widely used in pharmaceutical, biochemical, and research settings for compound identification and characterization.

How Does LC-MS Work?

In LC-MS analysis, a sample is first separated by liquid chromatography, which isolates individual components based on their chemical properties. The separated components then pass into a mass spectrometer, which measures the mass-to-charge ratio (m/z) of the ions produced from each component.

The resulting mass spectrum provides a highly specific "fingerprint" for each compound, allowing researchers to confirm the molecular identity of a substance with a high degree of confidence.

How Is LC-MS Used for Peptide Identity Testing?

For research peptides, LC-MS is used to confirm that the compound matches the expected molecular weight and fragmentation pattern of the target peptide. This identity confirmation is an important complement to HPLC purity testing, as it verifies not just that the sample is pure, but that the primary component is the correct compound.

Why Is LC-MS Important?

HPLC alone can confirm purity but cannot definitively identify what the compound is. LC-MS adds identity confirmation, providing a more complete picture of product quality. Together, HPLC and LC-MS data on a Certificate of Analysis give researchers greater confidence in the material they are working with.
